The methyltransferase Tgs1, a catalyst of TMG cap synthesis, is guanine-specific; N7 methylation must precede N2 methylation; it acts via a distributive mechanism; and the chemical steps of TMG synthesis do not require RNA or protein cofactors. [Tgs1]
