Data suggest that cdc48 cofactor ufd1 is required for SREBP1/2 cleavage/activation but does not interact with cdc48-rbd2 complex; cdc48-ufd1 is instead required at step prior to rbd2 function, during Golgi localization of dsc1-E3 ligase. (cdc48 = AAA family ATPase cdc48; ufd1 = cdc48-ufd1-npl4 complex component ufd1; dsc1-E3 ligase = ubiquitin-protein ligase E3; SREBP1/2 = sterol regulatory element binding proteins 1/2)
Title: Dsc E3 ligase localization to the Golgi requires the ATPase Cdc48 and cofactor Ufd1 for activation of sterol regulatory element-binding protein in fission yeast.
Dsc1 is an active E3 ligase; loss of Dsc E3 ligase activity leads to endoplasmic reticulum localization of the Dsc complex.
