Binding capacity was compared between StrepCaptureXP™ Resin Lite (GenScript, L01114) and a commercial affinity resin from Competitor-ST using six Strep-tagged proteins expressed in E. coli and CHO cells under identical purification conditions.

The target membrane protein (ASIC2a) has a theoretical molecular weight of 60.1 kDa.

Sample Preparation:
1. Cells were lysed in 20 mM HEPES, 150 mM NaCl (pH 7.4) containing 1–1.5% (w/v) LMNG for 2 h at 2–8°C.
2. The lysate was clarified by ultracentrifugation at 45,000 rpm for 45 min at 4°C.
3. 100 μL of resin was incubated with 12 mL of clarified membrane protein lysate for 2 h at 2–8°C.
4. After binding, washing and elution were performed according to the recommended purification protocol. Elution fractions were analyzed by SDS-PAGE.

StrepCaptureXP™ Resin Lite

Binding capacity of StrepCaptureXP™ Resin Lite (GenScript, L01114) was evaluated after repeated regeneration with 0.1 M NaOH. No significant loss in binding capacity was observed after 40 regeneration cycles.

GenScript’s StrepCaptureXP™ Resin Lite (Cat. No. L01114) is a next-generation affinity chromatography medium engineered for the efficient purification of Strep-tag® II and Twin-Strep-tag® fusion proteins. Functionalized with an innovatively engineered StrepCaptureXP ligand (a genetically optimized streptavidin variant), this resin enables high-affinity, high-specificity capture proteins from complex biological lysates, including those derived from mammalian, bacterial, and insect expression systems.
The resin enables gentle and reversible elution, helping to maximize the recovery of biologically active proteins. It is particularly suitable for applications requiring mild purification conditions, such as the purification of membrane proteins and transmembrane proteins.
