GenScript Protein A Resin is an affinity_x000D_
chromatography medium designed for easy, one-step purification of classes, subclasses_x000D_
and fragments of immunoglobulins from biological fluids and from cell culture_x000D_
media. Protein A Resin can also be used for immunoprecipitation of_x000D_
proteins, protein complexes or antigens. The recombinant protein A ligand is_x000D_
coupled to 4%_x000D_
agarose. The coupling is optimized to give high binding capacity for_x000D_
immunoglobulins. The static binding_x000D_
capacity of Protein A Resin is greater than 20 mg human IgG/ml settled resin._x000D_
The binding capacity will_x000D_
vary depending on several factors such as target antibody, flow rate etc. Table_x000D_
1 lists the characteristics of Protein A Resin. _x000D_ _x000D_ Protein A, a bacterial cell wall protein isolated_x000D_ from Staphylococcus_x000D_ aureus,_x000D_ binds to mammalian IgGs mainly through Fc regions. Native_x000D_ protein A has five IgG binding domains and many other domains with unknown_x000D_ functions. Recombinant protein A contains five high_x000D_ affinity IgG binding domains with other non-essential domains removed to reduce nonspecific binding. Since only the Fc region_x000D_ is involved in binding, the Fab region is available for binding antigens. |
High temperature heating is not recommended. The agarose melts above 65°C.
