GenScript offers hybridoma cell culture services for monoclonal antibody production in flexible scales, up to gram level. You send us your hybridoma cells. We carry out the hybridoma culture, antibody purification and deliver you the protein A/G purified antibody with tightly controlled endotoxin level and minimal to zero bovine Ig contamination. We guarantee the antibody yield from the hybridoma cell culture is 15-50 mg/L.

　 Description Time Line
Phase I: Cell line submission, recovery, adoption and expansion
 Customer submits:
1-2 weeks
2 vials of frozen hybridoma cells
The recipe of the culture medium for the submitted hybridoma cells
The name of the species from which the hybridoma cell line was derived
 The production history of the submitted hybridoma cell line (if available)
Once the above materials are received, the hybridoma cell line will be recovered and examined for any cell contamination. If there is no contamination, the hybridoma cell line will be adopted to production medium and expanded.
Phase II: Inoculation
 The expanded cell culture is then transferred into a roller bottle containing hybridoma cell culture medium to a final cell density of 1-2X105 cells/ml.
2 weeks
The hybridoma cells containing roller bottle is immediately placed on the roller device in a 37°C incubator
Hybridoma cells are then cultured under 2-3 rpm rotation for two weeks
Hybridoma cells are monitored daily for growth status during the hybridoma cell culture process
If needed, a large scale hybridoma culture can be done using the hybridoma cells from the existing roller bottle culture for further inoculation.
Phase III: Harvest & antibody purification
 hybridoma cell culture is halted by removing the roller bottle from the incubator and then stored at 2-8°C until all roller bottle cultures are completed.
1 week
The hybridoma cells are removed from the culture medium by centrifugation at 3,000-4,000 rpm for 15 min or by filtering through 1.0µm filters.
The culture supernatant is then concentrated to 1/10th of the original volume. The concentrated supernatant can be immediately processed for antibody purification. If not, the concentrated antibody-containing supernatant can be stored at -20°C.
Antibody is then purified by protein A/G and ready for delivery.
