e show that SPRTN is a DNA-dependent mammalian protease required for resolving cytotoxic DNA-protein crosslinks (DPCs)- a function that had only been attributed to the metalloprotease Wss1 in budding yeast. We provide genetic evidence that SPRTN and Wss1 function distinctly in vivo to resolve DPCs

Upon DNA damage Wss1/Cdc48/Doa1 ternary complex is recruited to sumoylated targets and catalyzes SUMO chain extension through a newly recognized SUMO ligase activity.

Data suggest that Wss1 contributes to survival of DNA-protein crosslink (DPC)-harboring cells by acting on DPCs proteolytically. Wss1 enables replication of DPC-containing DNA and thus promotes genome integrity.
